trastorno gen\u00e9tico poco frecuente que afecta principalmente a los varones. El s\u00edndrome est\u00e1 causado por una duplicaci\u00f3n de material gen\u00e9tico localizado en la regi\u00f3n terminal (Xq28) del cromosoma X que implica al gen MECP2 as\u00ed como a otros genes relacionados (<em>IRAQ1<\/em>, <em>GDI1<\/em>, <em>FLNA<\/em>, <em>L1CAM<\/em>, <em>IDH3G<\/em>\u2026). El tama\u00f1o de la duplicaci\u00f3n y la gravedad de los s\u00edntomas var\u00edan mucho de un paciente a otro, pero no existe una relaci\u00f3n estrecha entre estas estas dos variables.<\/p><p>La prote\u00edna codificada por el gen MECP2, denominada MeCP2 (methyl CpG binding protein 2) desempe\u00f1a un papel central en la regulaci\u00f3n de otras prote\u00ednas responsables del desarrollo y la funci\u00f3n cerebral.<\/p><p>La duplicaci\u00f3n del gen conduce a la sobreproducci\u00f3n de la prote\u00edna MeCP2, impidiendo as\u00ed la regulaci\u00f3n de otras prote\u00ednas. Esto se traduce cl\u00ednicamente en discapacidad intelectual (DI) de moderada a grave, retraso en el desarrollo psicomotor y lenguaje muy deficiente o inexistente.<\/p><p>Cuando las mutaciones conducen a la producci\u00f3n de una prote\u00edna MeCP2 no funcional, aparecen s\u00edntomas similares a los del s\u00edndrome de duplicaci\u00f3n MECP2. En m\u00e1s del 90% de los casos, la duplicaci\u00f3n se hereda de una madre portadora asintom\u00e1tica. El t\u00e9rmino portador se utiliza cuando la persona es portadora del defecto gen\u00e9tico pero este defecto no se manifiesta cl\u00ednicamente.\u00a0\u00a0<\/p><p>Durante un embarazo, hay un 50% de probabilidades de dar a luz a un ni\u00f1o afectado y un 50% de probabilidades de dar a luz a una ni\u00f1a con la mutaci\u00f3n. <span style=\"color: var( --e-global-color-text ); font-family: var( --e-global-typography-text-font-family ), Sans-serif; font-style: var( --e-global-typography-text-font-style ); font-weight: var( --e-global-typography-text-font-weight ); text-transform: var( --e-global-typography-text-text-transform ); background-color: var( --e-global-color-c15569b );\">Muy raramente las ni\u00f1as desarrollan una forma sintom\u00e1tica de la enfermedad.<\/span><\/p><p>Los varones (XY) s\u00f3lo tienen un cromosoma X. La duplicaci\u00f3n del gen MECP2 provoca s\u00edntomas graves en cualquier caso.<\/p><p>A diferencia de los hombres, las mujeres (XX) tienen dos cromosomas X. Una duplicaci\u00f3n del gen MECP2 afecta muy raramente a una mujer porque el cromosoma X con material duplicado est\u00e1 inactivo y el nivel de prote\u00edna MeCP2 permanece normal.<\/p><p>En raras ocasiones la mutaci\u00f3n se expresa en mujeres debido a una inactivaci\u00f3n desfavorable del cromosoma X mutado o porque la mutaci\u00f3n se transpone a otro cromosoma. En ambos casos, los s\u00edntomas van desde problemas psiqui\u00e1tricos (depresi\u00f3n y ansiedad) hasta s\u00edntomas graves id\u00e9nticos a los encontrados en los hombres afectados.<\/p><p>La duplicaci\u00f3n del gen MECP2 tambi\u00e9n puede producirse durante la fecundaci\u00f3n. est\u00e1 muy infradiagnosticado debido a la variabilidad de los s\u00edntomas, actualmente se desconoce la prevalencia exacta del s\u00edndrome y se estima que est\u00e1 presente en 1\/150.000 nacimientos masculinos.<\/p><p>En la actualidad se desconoce el n\u00famero de casos en todo el mundo. Se manifiesta principalmente en ni\u00f1as, con una incidencia de 1 de cada 10.000 a 15.000 nacimientos.
